Overview of Colistin
Colistin is a cationic polypeptide antimicrobial agent with activity against a range of Gram-negative organisms. It is a cyclopeptide antibiotic, also known as polymyxin E, produced as a fermentation product of Bacillus polymyxa var. colistin. Colistin is composed of at least 30 polymyxins, with the main fractions being colistin A (polymyxin E1) and colistin B (polymyxin E2), which differ only in the length of the acyl side chain. In addition to being used as an antibiotic for the treatment of multidrug-resistant gram-negative bacteria infections, colistin has also been widely used as a feed additive for the last several decades for its significant growth-promoting effect in livestock, which may lead to colistin residue in edible food such as milk and meat, and subsequently pose potential nephrotoxicity and neurotoxicity to human. Meantime, the low level of colistin in animal-origin food may also induce resistant bacteria as the predicted no-effect concentrations of colistin for resistance selection is only 2 ng/mL. Therefore, it is of great importance to developing measurement systems to monitor the colistin residue in animal-origin food for combating bacteria drug-resistance and ensuring public health.

Development of Colistin Hapten
As a small molecule that cannot elicit animal body to produce a specific antibody, colistin could be used as a hapten to linked with bovine serum albumin (BSA) and ovalbumin (OVA) through carbodiimide coupling method to form an immunogen and monoclonal antibodies were produced by immunizing the animal with it. Studies have shown that this mAb can specifically recognize colistin and it had negligible cross-reactivity with other antimicrobial drugs, such as bacitracin, aminoglycosides, β-lactams, tetracyclines, sulfonamides, quinolones, and chloramphenicol. With this specific monoclonal antibody, a rapid one-step ci-ELISA and an LFIA for colistin in animal feed and food were developed, which have been proved two convenient immunoassays to monitor the illegal use of colistin in animal feed and the colistin residue in animal-origin food.
