Introduction of Bacitracin

Bacitracin is an antibiotic isolated by the nutrient solution of Bacillus licheniformis or Bacillus subtilis mutation. With similar antimicrobial spectrum to penicillin G, bacitracin has a stronger restraining effect on gram-positive microorganisms and is mainly used in treating penicillin-fast staphylococcal infections. In addition, bacitracin has been also used as fodder additives in modern animal husbandry. As a consequence, residual bacitracin in milk and edible animal tissues might lead to the allergies and emergence of resistant microbes. Therefore, it is of great importance to detect bacitracin residues. Currently, detection methods such as immunoassays, liquid chromatography with ultra-violet, fluorescence detection, and mass spectrometry have been applied for the detection of bacitracin in commercial pharmaceuticals, feeds, and milk samples. Among these approaches, specific antigen-antibody interaction-based immunoassays represent a simple, accurate, and sensitive method that provides a means for rapid and efficient screening of the samples for bacitracin residue detection.

Development of Bacitracin Hapten

Designing of antibodies plays a very important role in the efficiency enhancement of the screening assay, especially in the antigen-antibody immunoreactive immunology-based approach. However, bacitracin is micromolecular compound that does not have immunogenicity, which means macromolecular carrier albumen must be coupled with bacitracin to obtain immunogenic artificial antigen and thus produce specific antibodies. It has been reported that bacitracin could be used as a hapten to prepare a complete antigen by coupling with common carrier albumen, such as bovine serum albumin (BSA), and ovalbumin (OVA), human serum albumin (HSA), mouse serum albumin (MSA), thyroprotein (TG) or hemocyanin (KLH), etc. Specific monoclonal antibody obtained by immunizing the animal with this antigen has been proved can be applicable to detect bacitracin with higher sensitivity.
