Peptides antibiotics, also known as antimicrobial peptides (AMPs), are short and generally positively charged peptides found in almost all organisms with remarkable structural and functional diversity. Studies have shown that many AMPs exhibit an extraordinarily broad range of antimicrobial activity covering both Gram-positive and Gram-negative bacteria as well as fungi, viruses, and unicellular protozoa. Moreover, several AMPs display the ability to modulate the innate immune responses of the host and thereby indirectly promote pathogen clearance. During the past, peptide antibiotics have been developed as novel anti-infectives to modulate the immune response, promote wound healing, and prevent post-surgical adhesions. Unfortunately, peptide antibiotics resistance is increasingly reported, leaving a serious threat to human health. Therefore, the establishment of rapid detection methods for PTs residues is highly essential to ensure public health.

Peptides Antibiotics (PTs) Hapten Development

To date, thousands of AMPs have been discovered. Most AMPs are relatively short, commonly consisting of 10-50 amino acids, display an overall positive charge ranging from +2 to +11, and contain a substantial proportion (typically 50%) of hydrophobic residues. Based on the different amino acid components and, these peptides can be classified into α-helical, β-sheet, or peptides with extended/random-coil structure, with most AMPs belonging to the first two categories. Common peptide antibiotics include actinomycin, bacitracin, colistin, and polymyxin B. To detect PTs residues, several PTs have been treated as haptens for specific antibodies preparation. For example, a novel antibody against colistin has been developed and applied to a series of immunoassays for colistin residues detection, such as ELISA kits development. Using this method, colistin-BSA (bovine serum albumin) and colistin-OVA (ovalbumin) conjugate are synthesized and used as immunogen to the immune animals. Moreover, combination hapten of both gentamicin and vancomycin with BSA has been synthesized to prepare antibodies for immunoassay used to detect gentamicin and vancomycin residues.
