Introduction to Rhodamine B

RB is a type of chemical compound that has been used as important fluorescent labeling for many immunoassays, such as flow cytometry (FCM) and enzyme-linked immunosorbent assay (ELISA). In general, the molecular formula of RB is C28H31ClN2O3 and its molecular weight is approximately 479.02 gm/mol. Meanwhile, pilot studies have indicated that RB can also be utilized as a colorant in many industries, including food and printing. However, recent reports have revealed that exposure to RB can cause a series of side effects to human safety and health. As a consequence, many attempts have been made recently to develop rapid and accurate methods for detecting RB or its derivatives in foodstuffs. For example, high-performance liquid chromatography (HPLC) is a common method for detecting RB in laboratories. Furthermore, a novel ELISA detection system for RB has been also generated by using new artificial antigen and small-molecule antibodies against RB analogs. The data have shown that these antibodies usually have high sensitivity to RB with an IC50 value of about 0.1-0.5 ng/mL and have great cross-reactivity to other food additives.

Our Platform for Design and Synthesis of Haptens for Rhodamine B

RB, a highly water-soluble tracer dry, has been broadly used as a food additive because of its low-cost and high stability. However, due to its carcinogenicity and mutagenicity, the determination of RB in different food samples is essential to human health and RB control.

Therefore, Creative Biolabs has developed an integrated RB hapten-based platform, ranging from RB hapten design and synthesis to RB hapten evaluation and modification. RB hapten will be generated by attaching to specific carrier proteins to trigger immune responses. Then, the rhodamine B artificial antigens will be developed by coupling RB or its derivatives with different types of proteins, including bovine serum albumin (BSA) and ovalbumin (OVA). Afterward, the antibodies against RB or its residues can be obtained by animal immunization. Eventually, the sensitivity and specificity of these antibodies can be fully assessed by using a panel of assays. Besides, we are capable of optimizing all the hapten design and synthesis workflows to reduce overall cost and shorten the development time.

With years of operational experience, Creative Biolabs provides state-of-art services for designing novel haptens for RB or its derivatives. Our scientists specialized in small-molecule antibody studies will work with you to establish the most appropriate strategy. Now, we have developed efficient protocols for hapten design and synthesis.
